YES, you can install the Jelly Bean update with the link above, even if your bootloader is unlocked. YES you can backup root with OTA RootKeeper ( and THEN install the Jelly Bean update, and restore root!
It’s HIGHLY recommended that you run NVFlash (Video here: BEFORE doing this update, because it will ENSURE that if you brick your device (ONCE UNLOCKED!!!), you’ll ALWAYS be able to restore it. A friend recently bricked his trying to install a ICS based Custom Recovery on a Jelly Bean rom, and it BRICKED his device! He didn’t use NVFlash, and Asus quoted him 240 dollars to repair his tablet! Another friend did something similar (Linked comment: and had to pay 225! MAKE SURE you run NVFlash is you EVER consider unlocking your Prime!!! This tablet is to expensive NOT TO do it, and I have an easy step by step video to go by as well….
